MDM2抗体（SMP14）是一种IgG1κ小鼠单克隆MDM2抗体（也称为HdmX抗体或HDM2抗体），可通过WB、IP、IF和IHC（P）检测小鼠、大鼠和人类来源的MDM2蛋白。MDM2抗体（SMP14）既可以是非偶联的抗MDM2抗体形式，也可以是多种偶联形式的抗MDM2抗体，包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。MDM，也称为鼠双微体2，最初被鉴定为小鼠转化系统中的癌基因。MDM2蛋白的功能是结合p53并阻断p53介导的共转染报告构建体的反式激活。在保留野生型p53的人肉瘤中，MDM2基因被高比例扩增，而过表达MDM2的肿瘤细胞可以耐受高水平的p53表达。这些发现表明，MDM2过表达至少代表了一种在肿瘤发生过程中p53功能被废除的机制。因此，MDM2抗体（SMP14）是癌症研究的重要试剂。p53是迄今为止人类癌症中最常见的突变基因。p53的表达通过阻止细胞从细胞周期的G1期进展到S期来抑制细胞生长。最重要的是，p53的功能是在细胞暴露于DNA损伤剂后，导致细胞在细胞周期的G1期被捕获。

SC-956 항체는 MDM2의 cleaved form만 확인할 수 있는 항체인가요? 저희가 상기 항체로 HepG2와 SNU398에서 확인하였을 때 데이타 시트에서 처럼 약 60 kd 위에서만 밴드가 확인되는데 맞는지 궁금합니다.

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue?

Thank you for your question. Yes. There are two options. You can use one of our Mouse IgG Binding Proteins as a highly specific secondary detection reagent. Alternatively, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal, MDM2 Antibody (SMP14): sc-965, is currently available conjugated to HRP (sc-965 HRP), phycoerythrin (sc-965 PE), fluorescein (sc-965 FITC), Alexa Fluor® 488 (sc-965 AF488), Alexa Fluor® 647 (sc-965 AF647), and biotin (sc-965 B).
