参与真核细胞有丝分裂调控的Cdc2/cyclin B酶受到多个层次的调控。其中，通过酪氨酸磷酸化对催化亚基的调控是最为了解的。酪氨酸磷酸化抑制Cdc2/cyclin B复合物，而酪氨酸去磷酸化在有丝分裂开始时发生，直接激活前MPH复合物。Cdc25基因作为限速的有丝分裂激活剂，显然是由于其作为Cdc2酪氨酸磷酸酶的作用。在缺乏Cdc25的情况下，Cdc2会累积在酪氨酸磷酸化的状态下。此外，来自多种物种的Cdc25蛋白已被证明与其他酪氨酸磷酸酶具有较低程度的序列相似性。Cdc25基因家族至少包含三个成员，在其最保守的羧基端序列中共享约40%的同源性。

On your images, I see that sometimes this antibody shows more than one band? Is it a phosphorylated or other isoforms or the other genes with ~40% similarity

Thank you for your question. The antibody does undergo a post-transnational modification which is phosphorylation. https://www.uniprot.org/uniprotkb/P30304/entry#ptm_processing

I am using Cdc25A (DCS-120): sc-56264 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
