Hi, I'm working on IF staining of YAP in fibroblasts, what fixation buffer do you recommend for that with YAP Antibody (G-6): sc-376830? And how about the cell confluence? Will lower or higher confluence influence the YAP location in cells?

Can this antibody be used in huaman BMMSCS samples in WB?

Thank you for your question. We have not tested this antibody specifically on Bone Marrow Mesenchymal Stem Cells, so I am unable to specifically say if the YAP protein is expressed in these cells. We recommend you research in the primary literature to see if these BMMSCS cells express the YAP protein.

Has anyone tried this anybody for ChIP? Our lab used the predecessor antibody (now discontinued) for ChIP and I need to do it again looking for some other binding sites.

Thank you for your question. We are not currently aware of any data or product citations using this antibody for ChIP research applications. We do offer an alternative antibody, YAP Antibody (63.7): sc-101199, which has been published in ChIP research applications.

Is antigen retrieval recommended for this antibody in IHC? If so, are there any guidelines about which antigen retrieval methods are best for the protein?

Hello, I'm interested in getting  a FREE 10 µg sample of YAP (G-6): sc-376830. We are working with the mouse intestinal organoids and would like to detect how YAP protein localisation changes during oncoprotein activation. I would appreciate your reply.

Can this mouse monoclonal antibody be used to stain mouse samples in IF and IHC?

Rated 5 out of 5 by from Nice and single band (w.b.)Produced a single band at the expected molecular weight (~65kDa). Sample: MDA-MB-231 wcl (30ug). Blocking: 1h in blocking buffer (0.05% tween-20 in TBS with 5% skin milk). Primary antibody: 1:200 in blocking buffer, overnight @ 4oC. Secondary antibody: 1:3000 in blocking buffer. Exposure time: 30 seconds* *further dilution of the primary antibody (1:400-1:500 ?) may yield similar results at the expense of a prolonged exposure time (2-4 minutes).
