TLR4抗体（25）是一种IgG1κ小鼠单克隆TLR4抗体（也称为CD284抗体或toll样受体4抗体），可通过WB、IP、IF和IHC(P)检测小鼠、大鼠、人类和犬类的TLR4蛋白。TLR4抗体（25）既可作为非缀合的抗-TLR4抗体形式，也可作为多种缀合形式的抗-TLR4抗体，包括琼脂糖、HRP。最初基于序列相似性，鉴定出了六种果蝇Toll受体的人类同源物，并将其命名为Toll样受体（TLR）。Toll受体参与介导发育中的果蝇胚胎的背腹极化，也参与宿主免疫。TLR蛋白家族的特点是具有高度保守的Toll同源（TH）结构域，这对于Toll诱导的信号转导至关重要。TLR1以及其他TLR家族成员都是I型跨膜受体，其特征是包含一个由几个富含亮氨酸的区域组成的细胞外结构域和一个单一的细胞质Toll/IL-1R样结构域。TLR2和TLR4在响应脂多糖（LPS）刺激时被激活，导致NFkB的激活和易位，这表明这些受体参与介导炎症反应。在免疫染色应用中，TLR4抗体（25）显示核和细胞质染色。TLR受体在外周血白细胞、巨噬细胞和单核细胞中的表达最高。TLR6与TLR1高度同源，序列同一性超过65%，并且与其他TLR家族成员一样，它在激活时诱导NFkB信号传导。

Buongiorno. Sarebbe possibile avere questo anticorpo coniugato per fluorescenza su fettine tessuto incluso in paraffina?

Thank you for your question. TLR4 Antibody (25): sc-293072 is not available with fluorescence conjugation. As possible alternatives we can offer TLR4 Antibody (HTA125): sc-13593, which is recommended for detection of TLR4/MD-2 complex of human origin by IP, IF, IHC(P) and FCM, or TLR4 Antibody (MTS510): sc-13591, which can be used for detection of TLR4/MD-2 complex of mouse, rat and human origin by IP, IF and FCM. Both are available with a variety of different conjugation options, which you can find on their individual product pages.

Is there any published paper about the IP application of this TLR4 antibody?

It is stated that this antibody is suitable for detecting TLR4 of mouse for WB. However, it is not stated that secondary antibody does not cross-react with mouse protein lysate. What is the recommended use of this antibody for WB from mouse tissue?

do you have a fluorescence tag version of TLR4 Antibody (25): sc-293072 available? or can you order a custom made conjugated version?

Thank you for your question. TLR4 Antibody (HTA125): sc-13593 and TLR4 Antibody (MTS510): sc-13591 are available in fluorescence-conjugated versions. We do not offer fluorescence conjugates of TLR4 Antibody (25): sc-293072.

Is there any chances of double bands in the immunoblot of mouse peritoneal macrophages cells lysates?

I didn't get positive result on mouse MC3T3E1 cells by using this TLR4 antibody with1:100 or 1:200 dilution,I was using 555 second antibody,what's the problem is?

I used a positive control, anti-MUC2, a sample previously and kindly provided by SC, which works very well. I do not exclude it may be a matter of low expression
