With what kind of magnetic beads these Abs work good for ChIP? I tried with Dynabeads Protein G and I didn't get it

Thank you for your question. We have not specifically tested this antibody using magnetic beads, and we do not sell those type of magnetic beads so we can not make a recommendation. This is a mouse monoclonal IgG1, so we would recommend asking the magnetic bead supplier for their recommendation. This specific monoclonal antibody has been validated for ChIP (please refer to the product citations for more details).

According to the info sheet, this antibody was RNAPII Ser5p specific, have you check the specificity against Ser2p/Ser7p or non phopsphorylated RNAPII?

Thank you for your question. This antibody is recommended for detection of hypo- and hyper-phosphorylated Pol II of mouse, rat and human origin.

Can I use this mouse monoclonal antibody for IHC or IF staining on mouse samples?

Yes, this Pol II Antibody (CTD4H8): sc-47701 is available conjugated to agarose (sc-47701 AC), 500 µg/0.25 ml agarose in 1 ml, for IP; HRP (sc-47701 HRP), 200 µg/ml, for WB, IHC(P) and ELISA; phycoerythrin (sc-47701 PE), fluorescein (sc-47701 FITC), Alexa Fluor® 488 (sc-47701 AF488) or Alexa Fluor® 647 (sc-47701 AF647), 200 µg/ml, for IF, IHC(P) and FCM.

Does the antibody specifically recognizes only Ser5-phosphorylated Pol II CTD? I would like to perform ChIP for total Pol II, as well as Ser5- and Ser2-phosphorylated Pol II. Would you recommend using this antibody as a control for total Pol II?

Rated 3 out of 5 by from Clean but low signalI got a trail size of this antibody and tested it on Murine erythroleukemia cells (MEL). I got clean sharp bands on Western blot, but the signal was very low. I didn't quantify the amount of protein, but the minimum lysate required to see a signal was roughly equivalent to that from 150,000 cells. IMO for Pol II that's a lot!
