PML蛋白是一种锌指转录因子，由于选择性剪接而表现为三种主要的转录产物。编码人类PML的基因位于染色体15q22。视黄酸受体α（RARα）基因的t（15；17）（q22；q11.2-q12）染色体易位几乎发生在所有急性早幼粒细胞白血病病例中，导致PML/RARα嵌合蛋白的表达。表达PML/RARα嵌合体的骨髓前体细胞不能分化，并因凋亡减少而表现出生长速度增加。PML/RARα通过招募核共抑制因子（N-CoR）-组蛋白去乙酰化酶复合物来转化骨髓前体细胞，该复合物对视黄酸依赖性骨髓分化至关重要。PML/RARα还招募DNA甲基转移酶，以诱导基因高甲基化和沉默，最终促进白血病的发生。

I am using PML (G-8): sc-377340 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
