Hello! Please tell me if p-Tyr Antibody (PY20): sc-508 is working in ELISA assay. If yes, could you recommend me a starting dilution range! I want to know if the p-Tyr Antibody (PY20) HRP variant is recommended for ELISA and in what dilution.

Thank you for your question. p-Tyr Antibody (PY20): sc-508 is recommended for detection of broad species by WB, IP, IF, IHC(P) and FCM. It is also available conjugated to HRP for ELISA. The starting dilution is 1:30, with a dilution range of 1:30-1:3000.

Which phosphotyrosine antibody is more sensitive in Western Blot: p-Tyr (PY99) or p-Tyr (PY20)? Which concentration is the recommended?

Thank you for your question. Both p-Tyr Antibody (PY20): sc-508 and p-Tyr Antibody (PY99): sc-7020 are featured in several hundred publications and are recommended for western blot and other applications. Both antibodies have a recommended starting dilution of 1:200 and work in a dilution range 1:100-1:1000. Either antibody is perfectly suited for western blotting, so you could order one or the other or get in touch with our technical service to arrange for free samples of the two antibodies.

for blocking, what would you recommend? Many people recommend 5% of BSA for phospho-protein..( should I stick with this rule.). but some phospho-protein can worked well with.. Skim milk (depends the antibody protocol..)

Thank you for your question. We recommend using 1% milk, 1% BSA in 0.05% Tween-20 for blocking and antibody incubations.

We recommend using a directly conjugated mouse monoclonal primary antibody. See p-Tyr (PY99): sc-7020 for p-Tyr antibody conjugates, including HRP, FITC, PE, Alexa Fluor® 488 and Alexa Fluor® 647. https://www.scbt.com/scbt/product/p-tyr-antibody-py99
