IDH1 (F-3) 是一种小鼠单克隆IgM kappa抗体（也称为IDH抗体、PICD抗体或草酰琥珀酸脱羧酶抗体），可通过蛋白质印迹（WB）、免疫沉淀（IP）、免疫荧光（IF）和酶联免疫吸附测定（ELISA）检测小鼠、大鼠和人类来源的IDH1蛋白。IDH1 (F-3) 抗体可作为非结合型抗 IDH1 抗体使用。异柠檬酸脱氢酶（IDHC 或 IDH）是一种细胞质酶，属于异柠檬酸和异丙基丙二酸脱氢酶家族，由 416 个残基组成。IDHC催化柠檬酸循环的第三步，即异柠檬酸的氧化脱羧，在两步反应中形成α-酮戊二酸和二氧化碳。第一步是将异柠檬酸氧化为中间产物草酰琥珀酸，第二步是生成α-酮戊二酸。在此过程中，NADH或NADPH与二氧化碳一起生成。Ca2+可以与IDHC结合，形成异柠檬酸复合物，从而成为Mg2+的竞争性抑制剂。IDHC酶在Ser 113处被磷酸化而失活，并包含一个类似扣环的结构域，其中二聚体中的两条多肽链相互连接。IDHC在多种物种中表达，也存在于缺乏完整柠檬酸循环的生物中。

Thank you for your question. This antibody is provided as 200 µg in 1 ml PBS with 0.1 % gelatin and <0.1 % sodium azide.

To verify, this antibody recognizes the wild type IDH-1 and not the mutated version correct? I had read a publication where the investigators used a Santa Cruz antibody for the wild type and it was a goat-anti-human which is no longer available. Thank you

Thank you for your question. This monoclonal antibody is developed to recognize the wild type IDH1 protein. We have not specifically evaluated the antibody for potential cross-reactivity with the mutated IDH1 protein, which results from a mutation in Arg-132 that are tissue-specific. The antibody is raised against an epitope mapping between amino acids 33-58 near the N-terminus of IDH1.

Rated 1 out of 5 by from Unable to detect IDH1 in cells overexpressing IDH1We have attempted to use this antibody multiple times to detect IDH1 in cells designed to overexpress IDH1 or IDH1 mutant. At best, we detect a very faint band; typically, we see no band at all. Would love to know if we could do something to improve this.
