c-Fos抗体（D-1）是一种IgG2b κ型小鼠单克隆抗体，可通过WB、IP、IF、IHC(P)和ELISA等方法检测来自小鼠、大鼠和人的c-Fos蛋白。c-Fos抗体（D-1）既有偶联形式，也有非偶联形式。c-Fos原癌基因最初是在两个独立的小鼠骨肉瘤病毒分离株和一个禽类肾母细胞瘤病毒中检测到的。其细胞同源物c-Fos编码一种核磷酸化蛋白，可被多种因子快速且短暂地诱导，并作为几种基因的转录调节因子。与形成同型和异型二聚体并与特定DNA应答元件结合的c-Jun蛋白不同，c-Fos蛋白仅作为与Jun基因家族成员形成的异型二聚体时才具有活性。c-Fos的功能同源物包括Fra-1、Fra-2和Fos B基因。此外，选定的ATF/CREB家族成员可以与Fos和Jun形成亮氨酸拉链二聚体。不同的二聚体对AP-1和CRE位点表现出不同的特异性和亲和力。

what is the best replacement for Sc-52

I used immunofluorescence staining in the hypothalamus of rat, and as a result c-fos was expressed in the cytoplasm. It's not expressed in the nucleus. Is it correct? 0 answers

Thank you for your question. According to the Human Protein Atlas, c-Fos (FOS) is localized to the nucleoplasm. Nuclear and cytoplasmic expression may be detected in most tissues. https://www.proteinatlas.org/ENSG00000170345-FOS/pathology

Can this antibody be used for the detection of c-fos in the macaque monkey. If not, which of your antibodies would you recommend?

Thank you for your question. We have not tested this antibody for reactivity with macaque, and are not aware of any publications citing use of the antibody in that species. We do offer an alternative antibody, c-Fos (6-2H-2F): sc-447, that has been published at least twice for detection of c-Fos in monkey. Please see PMID's: 9108029 and 11937718 for those references. Could you please advise if this antibody is able to detect Fra proteins as is the case with the discontinued sc 253.

Thank you for your question. The equivalent replacement mouse monoclonal antibodies to sc-253 are sc-166940: c-Fos (E-8) and sc-271243: c-Fos (C-10). Both of these mouse monoclonal antibodies are raised against the same peptide antigen used to generate the polyclonal antibody, sc-253. These recommended replacement mouse monoclonal antibodies are also recommended for detection of c-Fos, Fos B, Fra-1 and Fra-2 of mouse, rat, human, zebrafish and avian origin. Rated 3 out of 5 by from Nucleus hypocampus mark but not hypothalamusWe tested this antibody 1:1000 in cryosectioned rat brains. Strong nuclear staining is observed in the hypocampus but not in the suprachiasmatic nucleus (SCN) where there should be staining since these animals were euthanized at the begining of their resting phase. Using cFos antibodies from other companies we were able to acquire a positive staining for the SCN. This antibody will not be usefull for us since it is not comparable with IHC using other antibodies.

Rated 1 out of 5 by from Not satisfied!I tried a free sample of this antibody on free floating mouse brain sections and it didn't work. I used a concentration of 1:50 for the primary antibody, and 1:300 for secondary antibody (anti-mouse 488) and I didn't got any signal. Really not satisfied.
