Is there anybody know the difference between PY20 and PY99?

Thank you for your question. p-Tyr Antibody (PY99) is recommended for detection of phosphotyrosine-containing proteins by WB, IP, IF and IHC(P); non cross-reactive with phosphoserine or phosphothreonine. p-Tyr Antibody (PY20) is recommended for detection of broad species by WB, IP, IF, IHC(P) and FCM.

Can I use 10% milk to block when using this antibody? I am interested in getting it conjugated with the alexa 790 to be used with LI-COR®Odyssey®.

I want to measure insulin sensitivity and several tyrosine domains are phosphorylated in response to insulin. For example in both insulin receptors and in IRS. Does the antibody detetct all p-tyr, and I differentiate between them depending on weight?

Yes, you are correct, this mouse monoclonal antibody, p-Tyr (PY99): sc-7020, specifically detects phosphotyrosine-containing proteins (and does not cross-reactive with phosphoserine or phosphothreonine-containing proteins). If you are using the antibody for Western blotting, then the molecular weight you detect in your samples is representative of different phosphorylated proteins. You would need to do further testing to identify which specific proteins are phosphorylated in your samples.
