카스파제-3 항체(31A1067)는 마우스, 쥐 및 인간 유래의 카스파제-3 및 전장 프로카스파제-3를 WB, IP, IF, IHC(P) 및 ELISA로 검출하는 IgG1 마우스 모노클로날 카스파제-3 항체입니다. 카스파제-3 항체(31A1067)는 비접합 항카스파제-3 항체 형태로도 제공됩니다. 아포파인, SCA-1, 야마, CPP32로도 알려진 카스파제-3는 카스파제의 ICE 하위 계열에 속하는 아스파테이트 특이적 시스테인 프로테아제입니다. 카스파제-3는 세포에서 비활성 전구체로 발현되며, 이로부터 성숙한 카스파제-3의 p17 및 p11 하위 유닛이 세포 사멸 중에 단백질 분해적으로 생성됩니다. 카스파제-3 전구체는 먼저 Asp175-Ser176에서 절단되어 p11 서브유닛과 p20 펩타이드를 생성합니다. 그 후, p20 펩타이드는 Asp28-Ser29에서 절단되어 성숙한 p17 서브유닛을 생성합니다. 활성 카스파제-3 효소는 2개의 p17과 2개의 p11 서브유닛으로 구성된 헤테로다이머입니다. 세포 사멸이 시작되면 카스파제-3는 Asp216-Gly217 결합에서 PARP를 단백질 분해적으로 절단합니다. 세포 사멸 캐스케이드가 실행되는 동안 활성화된 카스파제-3는 정상적인 스테롤 의존적 활성화와는 구별되는 단백질 분해 반응을 통해 ER 막에서 SREBP를 방출합니다. 카스파제-3는 기본 나선 루프-헬릭스 류신 지퍼 도메인과 막 부착 도메인 사이에서 SREBP를 절단하고 활성화합니다. 카스파제-3는 또한 카스파제-6, -7 및 -9를 절단하고 활성화합니다. 인간 카스파제-3 유전자는 폐, 비장, 심장, 간, 신장 및 면역계 세포에서 많이 발현되는 세포질 단백질을 암호화합니다.

I could not detect specific band on following condition on my western blot. ･Nitrocellulose membrane 0.45 µm ･C2C12 myoblast and C2C12 myoblast + H2O2 (500mM) ･Caspase-3 antibody: 1:200 ･IgGk antibody: 1:1000 Could you please give me some advice?

Can you recommend a compatible diluent buffer and concentration for this antibody? My blot showed up with a strong background and I was wondering what need to be modified?

Thank you for your question. This antibody was established using UltraCruz® Blocking Reagent: sc-516214, which is a highly efficient plant protein based blocking buffer. You may also want to increase incubation time of the primary antibody to overnight incubation at +4° C, which allows for lowering the overall primary antibody concentration, and in consequence unspecific signals. If the issue persists, you may also want to consider using one of our secondary binding proteins, which circumvent unspecific signals from secondary detection in comparison to conventional polyclonal secondaries.
