pADPr Antibody (10H) は IgG3 マウスモノクローナル pADPr 抗体（Poly(ADP-ribose)抗体、PAR 抗体、ADP-ribose polymer 抗体とも呼ばれる）で、WB、IF、ELISA で pADPr タンパク質を検出します。ポリ(ADP-リボシル化)は DNA 損傷に依存した翻訳後修飾の一つであり、傷害を受けた増殖細胞を細胞死から救うのに役立っている。PARP（ポリ[ADP-リボース]ポリメラーゼ）タンパク質は、ヒストンや他の核タンパク質を機能的に修飾し、それによって細胞死を防ぐ酵素のスーパーファミリーを構成する。PARPはNAD+を基質として、ADP-リボース残基をタンパク質のアクセプターに触媒的に転移する。この過程を何度も繰り返すと、ポリ（ADP-リボース）（pADPr）鎖が形成され、この鎖は独立して存在するか、標的タンパク質に結合する。この鎖が存在すると、標的タンパク質の機能が変化し、細胞の生存が促進される。さらに、pADPr鎖は細胞周期の進行やDNA損傷に対する細胞応答に重要であると考えられている。

Is this antibody suitable for immunoprecipitation studies?

Thank you for your question. We haven’t tested this antibody for immunoprecipitation (IP) and so that is not a recommended application at this time. How much sample should be loaded on SDS-PAGE for detecting pADPr using this antibody?

What is the most recommended diluent for this primary antibody? Is it ok to use 5% BSA in the solution? Thank you in advance.

Thank you for your question. We recommend diluting the primary antibodies in 5% milk with TBS-T. It is OK to use BSA, but we test them using milk as it tends to give cleaner results. Please contact technical support via phone, email or chat if you have any further questions!

I am all out of my donkey anti-mouse secondary antibody, what should I use instead?

によって、 5つ星のうち 4と評価されています。 It works for western blotting!It detected high molecular size bands, as expected for poly-ADP-ribosylated proteins, in total nuclear protein lysates from HEK 293T cells (1:500 dilution in 5% milk in TBS/0.1% Tween-20 overnight at 4C). Though, I recommend checking antibody specificity with PARP1-specific siRNAs or pharmacological inhibitors.

I used it on both Histochoice and 4% PFA fixed fibroblasts, and used it at dilutions of 1:200, 1:100, and 1:50, and even tried a 1hr DAKO antigen retrieval incubation at 95C, after speaking to technical support, all without being able to get any specific signal.
