I am using Vimentin (5G3F10): sc-66002 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 5 out of 5 by from Versatile and High-Performing Vimentin AntibodyI used this Vimentin antibody in both breast cancer and choriocarcinoma cell models, and it performed exceptionally well across multiple applications. In Western blotting and immunoblotting, it produced clear, specific bands with minimal background (optimal starting dilution: 1:200; range: 1:100–1:1000). For immunoprecipitation, using 1–2 μg of antibody per 100–500 μg of total protein (1 ml lysate) yielded efficient pulldown with strong specificity. In immunofluorescence assays, the antibody produced crisp, distinct staining with a starting dilution of 1:50 (range: 1:50–1:500). It also worked very well in immunohistochemistry, including paraffin-embedded sections, delivering robust staining patterns at similar dilutions. Overall, this is a reliable, multi-functional antibody suitable for a wide range of experimental approaches.

Rated 4 out of 5 by from Specificity was confirmed by WT and Vim-/- MEFsThe specificity of the antibody was confirmed by the WT and Vimentin knockout mouse embryonic fibroblasts (MEFs), but the staining pattern, which is typical of monoclonal antibodies against the cytoskeleton, showed a strong dotted staining only where the epitope was exposed.
