Hi, I would like to know if this antibody (sc-5277 AF488) can be used for flow cytometry? At which concentration I should use it for flow cytometry and for western blot ? Thanks

Thank you for your question. Fluorescent conjugates of this antibody can be used in flow cytometry, where adding 1 µg of fluorochrome-conjugated antibodies to tubes containing 100 µl of cell suspension (equal to 1 million cells). The unconjugated or HRP-conjugated antibody has a recommended starting dilution of 1:200 for western blots, with a dilution range of 1:100-1:1000.

Does it recognize mouse TCR beta or only human

I tried this Ab for IF with dilution 1:50 on 4% PFA fixed Jurkat cells but I didn't see the specific staining. How should I troubleshoot it?

Any idea how much to use TCR β Antibody (G-11) for Flow Cytometry? I tested at final staining concentration 5ug/ml and failed to see any positive stain in purified human T-cells.

Why is TCR β (G-11): sc-5277 the recommended alternative to the now discontinued TCR β (H-197): sc-9101?

Thank you for your question. The monoclonal primary antibody TCR β (G-11): sc-5277 offers a higher degree of specificity and essentially eliminates all lot-to-lot variation. It is also warrantied for all of the same applications as the discontinued polyclonal antibody: WB, IP, IF, and IHC(P).
