Is this antibody reactive to mouse cells/tissues?

Can SQSTM1 Antibody (D-3) be done on frozen tissue? What protocol will you recommend ?

Thank you for your question. Anti-SQSTM1/p62 Antibody (D-3): sc-28359 is recommended for detection of SQSTM1 of human origin by WB, IP, IF, IHC(P) and ELISA. The recommendation for IHC staining covers both paraffin-embedded and frozen tissue sections. I work with porcine brain capillary endothelial cells. Has this antibody been tested in pig or porcine cells?

Thank you for your question. SQSTM1 Antibody (D-3): sc-28359 has been tested in cells and tissues from mouse, rat, and human and is only recommended for detection of sequestosome 1 of human origin by WB, IP, IF, IHC(P) and ELISA.

Are there supplied data that measured expression in mouse liver? When I check the expression of SQSTM1 at mouse liver, SQSTM1 band is formed at about 55kda. I think it is not 62kda. So I want to see supplied data in mouse liver if you have.

Thank you for your question. The expected molecular weight for SQSTM1 is closer to 62kDa, but there are many reasons why it can run slightly lower. Do you have any suggestions for the dilution and incubation time for use of SQSTM1 Ab (D-3) in IHC on paraffin embedded tissues?

Rated 4 out of 5 by from Fine for WB, good for IPI have tested this antibody in quite a few western blot in MCF7 cells. In this cell line, it generates a few bands but a (very) major one at the expected molecular weight, which is reduced upon treatments promoting autophagy, which further authenticates the band. It works pretty well for immunoprecipitation.

Briefly - 30 micrograms of total protein extracts from human mesothelioma cells were separated by SDSPAGE; - the 0.45 nitrocellulose blot was then incubated 1h a RT with the SQSTM1 (D-3): sc-28359 Ab diluted 1:500 in 1X TBS 5% BSA, 0.02% NaAzide, phenol red followed by - Anti-Mouse 1:5000 in 5% milk in 1X TBS for 1h a RT -Detection through 'Immobilon ECL Ultra Western HRP Substrate ' after 30sec exposure
