WHICH ANTIGEN RETRIEVAL SOLUTION IS RECOMMENDED AND WHICH TISSUE SHOULD BE USED AS POSITIVE CONTROL

Thanks for your question. We recommend using sodium citrate with heat for antigen retrieval for this Sox-2 antibody. Here is a link to the recommended protocol: https://www.scbt.com/resources/protocols/immunoperoxidase-staining Suitable positive control tissues would be bronchus tissue or brain tissue.

1) Can this Sox2 antibody (SC365964) be used for IHC-frozen sections? 2) There are several different versions (sc365964, sc365964 X, etc.). How do I know which one to choose? I want a non-conjugated antibody for IHC.

Thank you for your question. We do recommend this antibody for IF, or IHC on frozen tissue sections. If you want a non-conjugated antibody, we would recommend purchasing sc-365964. The “X” product is more concentrated for ChIP and EMSA, and the other forms are all different conjugates.

Can the TransCruz product sc-365964 X be used for applications other than ChIP?

Thank you for your question. Yes, the TransCruz sc-365964 X can be used for other listed applications such as Western Blot or IF, the dilution just needs to be adjusted for the fact that the X form is ten times as concentrated as the standard antibody.

SOX2 is a transcription factor that is best known for its role in maintaining the pluripotency of stem cells however, in diffuse gliomas, including GBM, IHC for SOX2 shows remarkable specificity for infiltrating glioma cells, while non-neoplastic cells remain largely negative. Example of IHC staining for SOX2 show that only neoplastic cells exhibit significant nuclear staining.
