PTP1B抑制剂可抑制蛋白酪氨酸磷酸酶1B的活性,而蛋白酪氨酸磷酸酶1B是胰岛素和瘦素信号通路负调控的关键因素。分子动力学研究表明,由于在活性位点周围形成了氢键网络,PTP1B抑制剂具有强结合和高选择性。如果PTP1B在CHO和HEK293稳定细胞克隆中过表达,则在整个克隆生成和维持过程中会导致细胞毒性低和复苏率低。研究表明,PTP1B抑制剂可以显著放大PTP1B过表达的CHO细胞中关键葡萄糖转运蛋白Glut4的膜转位。PTP1B抑制剂选择性增加胰岛素受体、胰岛素受体底物1和Akt的磷酸化,模拟胰岛素的作用。PTP1B抑制剂也称为蛋白酪氨酸磷酸酶1B抑制剂,化学名称为3-(3,5-二溴-4-羟基苯甲酰)-2-乙基-N-[4-[(2-噻唑基氨基)磺酰基]苯基]-6-苯并呋喃磺酰胺。

What is the IC50 of this product? The website has two different numbers and I am not sure what they designate.

Thank you for your question. This chemical, PTP1B Inhibitor: sc-222227, is a non-competitive allosteric inhibitor of PTP1B (IC50 = 4 µM and 8 µM for PTP1B403 and PTP1B298, respectively). There are two IC50 values because the inhibitory concentration was measured at two different (assumed) amino acids.

(PubMed 26337385) report that Mena associates constitutively with the tyrosine phosphatase PTP1B and mediates a novel negative feedback mechanism that attenuates receptor tyrosine kinase signaling.
