What anti BDNF antibody to use for capturing mature BDNF using Immunoprecipitation? Previously (N-20): sc-546 antibody was used for the same but it has been discontinued.

is it possible to do IHC on 4% PFA fixed mouse brain with this antibody? Or is the sc-65513 possible?

We have not yet tested either pro BDNF antibodies (sc-65514 and sc-65513) on formalin-fixed paraffin embedded tissue sections and so IHC(P) is not a recommended application at this time. Check back for updates!

Can pro BDNF (5H8): sc-65514 monoclonal antibody be used for detecting mature BDNF?

Thank you for your question. This antibody, pro BDNF (5H8): sc-65514, is not recommended for detection of mature BDNF. The antibody recognizes an epitope that is specific for detection of BDNF preproprotein.

Rated 2 out of 5 by from Rather so-soIn immunohistochemical staining (rat brain tissue fixed in formalin), the antibody produced rather disappointing results. Cell bodies were left unstained, and only cellular processes, not sure whether specifically, were stained. Numerous dilutions, procedures, and antigen retrieval techniques were examined, also taken from cited publications. Current rating: 2 out of 5, but there are still ongoing optimisation efforts.

Rated 5 out of 5 by from mouse anti-proBDNF antibody (sc-65514)I have performed immunohistochemistry at a 1:100 dilution of your mouse anti-proBDNF antibody (sc-65514) and have good images to include in the datasheet. The tissue is mouse brain (prefrontal cortex and dorsal hippocampus) and has been perfused in 4% paraformaldehyde and cut by vibratome at 40 microns of thickness. DAB has been used as a chromogen.

Le prove hanno mostrato segnale aspecifico in particolare a livello del giro dentato dell'ippocampo. Lo stesso anticorpo è stato testato anche mediante western blot (1:500) su omogenati di encefalo di bovino. Le prove hanno mostrato segnale aspecifico con bandeggio sopra i 50 KDa in tutte le aree cerebrali analizzate (ippocampo, corteccia frontale, c. temporale e cervelletto) e nessun segnale sotto i 50 KDa.
