Periostin抗体（F-10）是一种IgG1κ小鼠单克隆Periostin抗体（也称为POSTN抗体、OSF2抗体、α-V/β-3和α-V/β-5整合素配体抗体、维生素K依赖性上皮细胞粘附和迁移因子抗体、分泌性细胞外基质蛋白抗体或伤口愈合/心室重塑相关蛋白抗体），可通过WB、IP、IF、IHC(P)和ELISA检测小鼠、大鼠和人类来源的Periostin蛋白。Periostin抗体（F-10）既有非偶联抗Periostin抗体形式，也有多种偶联形式的抗Periostin抗体，包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。Periostin（PN），也称为成骨细胞特异性因子2（OSF-2），是一种二硫键连接的蛋白，最初被分离为成骨细胞特异性因子。Periostin是一种分泌性蛋白，可与肝素结合，并作为α(V)β(3)和α(V)β(5)整合素的配体发挥作用。在成骨前细胞中，Periostin作为细胞粘附分子发挥作用，并参与成骨细胞的募集、扩散和附着。Periostin主要在下胃肠道、主动脉、胃、胎盘、子宫和乳腺组织中检测到，但在上皮性卵巢肿瘤中上调，在乳腺癌中过度表达。骨形态发生蛋白（BMP2）和转化生长因子β1（TGF β 1）可增加Periostin的表达。Periostin包含一个典型的信号序列，后跟一个富含半胱氨酸的结构域，一个四倍重复的结构域，该结构域与昆虫蛋白fasciclin具有同源性，以及一个C端结构域。

Hello. I used this antibody for western blot. However, the periostin was detected at about 70kda. Is it possible? Cell lines were HepG2 and LX-2.

Thank you for your question. Periostin has been reported to be expressed as many different isoforms. The full length protein is often described in the literature as a 90 kDa secreted glycoprotein. The literature has also described isoforms at ~84 kDa and 74 kDa with cleavages in the C-terminal region (CTR), see PMID: 19887451 as a reference.

Can Periostin (F-10): sc-398631 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Periostin (F-10): sc-398631 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
