p-GSK-3α抗体(E-2)是一种IgM κ小鼠单克隆p-GSK-3α抗体(也称为p-GSK-3α抗体或GSK3抗体),可通过WB、IP、IF和ELISA检测小鼠、大鼠和人源Ser 21磷酸化的GSK-3α。p-GSK-3α抗体(E-2)可以以非偶联的抗p-GSK-3α抗体形式提供。糖原合成酶激酶-3α(GSK-3α)和GSK-3β是丝氨酸/苏氨酸激酶的高度相似异构体,调节代谢酶和转录因子,负责协调糖原合成和细胞粘附等过程。GSK-3β的活性也是Rel二聚体核活性所必需的,Rel二聚体介导小鼠中TNFα的抗凋亡应答。GSK-3催化激酶活性通过丝氨酸/苏氨酸残基的差异磷酸化来控制,具有抑制作用,而酪氨酸残基具有激活作用。生长因子刺激表达GSK-3α和GSK-3β的哺乳动物细胞分别通过磷脂酰肌醇3-激酶(PI 3-K)-蛋白激酶B(PKB)依赖性途径诱导Ser 21和Ser 9的磷酸化,从而增强增殖信号。此外,GSK-3与cAMP依赖性蛋白激酶A(PKA)物理性结合,后者使GSK-3α的Ser 21或GSK-3β的Ser 9磷酸化并使两种形式失活。GSK-3α/β分别受Tyr 279和Tyr 216磷酸化的正调控。活化的GSK-3α/β参与能量代谢、神经元细胞发育和身体模式形成。GSK-3的酪氨酸去磷酸化参与其细胞外信号依赖性失活。
