Has this antibody been raised against the native or denatured structure of the protein? Is this information available somewhere for all anitbodies?

Thank you for your question. sc-376751 was ran under denaturing conditions. If an antibody is ran under reducing conditions it will be listed on the product page .

Hello, I am going to try WB for e-NOS- dimers and monomers. I found the article where they used sc-654 NOS3 Abs. Then I found that it has been replaced on sc-376751.Does it work the same? May I have a sample? Any suggestions for using LT-PAGE procedure?

Thank you for your question. Both the discontinued antibody and its successor have been raised against C-terminus of the protein. However, since the former is a polyclonal antibody, they are not identical. NOS3 Antibody (A-9): sc-376751 is recommended for detection of NOS3 of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. Does this Ab detect Ser-1177 phosphorylated form of nos3?

Thank you for your question. This antibody has not been epitope mapped, but it does have an epitope surrounding Ser 1177. As long as the exact epitope isn't in the location of the phosphorylation, it will be able to detect both non-phospholylated and phosphorylated forms.

Does sc-376751 work for IF for mouse tissue? If it does, what dilution would you suggest?

Thank you for your question. Yes, this antibody, sc-376751, NOS3 (A-9), is mouse reactive, and is recommended for IF. We recommend a starting dilution of 1:50 for IHC and IF use.

what is the difference between NOS3 Antibody (A-9): sc-376751 and NOS3 Antibody (C-6) sc-376542 in immunohistochemical staining of cardiac tissue in rats and what is the recommended positive control and how antigen retrieval is performed?

Thank you for your question. These are very similar monoclonal antibodies for NOS3 that recognize distinct epitopes. We do not have any data specifically in rat cardiac tissue. However, the sc-376751: NOS3 (A-9) antibody has been published in rat cardiac samples several times for Western blotting, IF and IHC. For example, please see PMID: 29333212 and 30120384. We have used FFPE human pancreas and spleen as positive controls for IHC(P). The recommended antigen retrieval method was sodium citrate buffer (pH 6) with heat treatment. Can you recommend a positive control to use with sc-376751: NOS3 (A-9) for Western blot?

We recommend A-673: sc-2414, MCF7:sc-2206 or HUV-EC-C: sc-364180 whole cell lysates as we have great Western blot data showing specific detection of myogenin in these cell lines.

Rated 5 out of 5 by from niceWe used this antibody to detect NOS3 in brain tissue of mouse by Western blot analysis and the results are amazing. If you use SDS-PAGE gel, we suggest that run the gel for 0.5 h at 40 V, then turn to 100V for 1-1.5h, you will get nice blot.
