Is it possible that NOS1 have strong expression in the nucleus of neurons?

Does this antibody works for zebrafish?

Please contact our Technical Service Department if you would like help performing sequence analysis with this antibody's antigen and the NOS1 protein sequence for zebrafish to predict potential cross-reactivity.

I used this antibody to stain nNOS of mouse origin and it worked quite well. But can this antibody recognize both splicing variants, nNOSalpha and nNOSbeta of mouse origin?

Thank you for your question. The exact epitope recognized by this mouse monoclonal antibody, NOS1 (A-11): sc-5302, has not been determined. The antibody is raised against amino acids 2-300 of NOS1 of human origin. According to Uniprot, mouse isoform nNos beta is missing amino acids 1-230 (http://www.uniprot.org/uniprot/Q9Z0J4#sequences). Therefore, it is possible that this antibody could recognize both variants.

Rated 1 out of 5 by from NOS1 in pig gutAt necropsy, intestinal tract of pig were dissected. Sample was washed in NaCl 0.9% and post-fixed by immersion for 7 h in 10% buffered formalin (pH 7.4), dehydrated through a graded series of ethanol and finally embedded in Paraplast X-TRA. Four-µm thick sections were cut in the transversal plane and collected on silane-coated slides. After deparaffinization and rehydratation, the Avidin/Biotin blocking was performed and slides were then incubated overnight at room temperature in a dark moist chamber with anti-NOS1 (A-11) (sc-5302) diluted 1:100 in phosphate buffer saline (PBS). After washing in PBS, sections were incubated for 1.5 hour with Goat Anti-Mouse IgG Antibody (H+L), Biotinylated (6 μg/ml, Vector Laboratories Inc., Burlingame, CA, USA) in a dark moist chamber. The sections were washed twice in PBS and treated with Fluorescein–Avidin D (Vector Laboratories Inc., Burlingame, CA, USA). Finally, the slide with tissue sections was embedded in Vectashield Mounting Medium with DAPI (H-1200, Vector Laboratories Inc., Burlingame, CA, USA) and observed using a Confocal Laser Scanning Microscope (FluoView FV300, Olympus, Tokyo, Japan). Our results did not demonstrate any NOS1-immunopositivity in any region of the pig gut.

The antibody worked well with mice but not hamster samples. The condition I used was 1:100 dilution. The cells were fixed with 4% PFA.
