Does this antibody stain all macrophages, meaning both M1 and M2? Thank you very much

Thank you for your question. This Macrophage Marker Antibody (MAC387): sc-66204 does not stain all macrophages, but a subset of macrophages (PMID: 3309045).

When probing tissues with this antibody, is it important that the tissues first be perfused and then post-fixed before embedding in OCT, or can I harvest the tissue, immediately snap-freeze in OCT, and then post-fix after cutting the tissue?

Thank you for your question. Frozen tissue blocks may be stored at -70° C until ready for sectioning. Thaw slides at room temperature prior to fixing and staining. Does this ab distinguish between suspension U937 cells and U937s that have been differentiated into macrophages? Thanks

Thank you for your question. We are not aware of any publications citing use of our Macrophage Marker Antibody (MAC387): sc-66204 in U-937 cells so we can not answer this question. The MAC387 clone recognises the L1 or Calprotectin molecule, an intracytoplasmic antigen comprising of a 12kD alpha chain and a 14kD beta chain expressed by granulocytes, monocytes and by tissue macrophages.

what is the kDa of this product sc-66204?

Thank you for your question. This Macrophage Marker Antibody (MAC387) is a mouse monoclonal recommended for detection of macrophages of mouse, rat and human origin. There is no standard expected molecular weight to be observed in kDa when using this antibody in Western blot, because the antibody does not detect a specific protein of a single size. Instead, it detects an antigen present in all macrophages.
