LAT1抗体(D-10)是一种IgG1κ小鼠单克隆LAT1抗体(也称为溶质载体家族7成员5抗体、溶质载体家族7(氨基酸转运蛋白轻链L系统)成员5抗体、大型中性氨基酸转运蛋白小亚基1抗体、L型氨基酸转运蛋白1抗体、CD98轻链抗体、CD98抗体、溶质载体家族7(阳离子氨基酸转运蛋白Y+系统)成员5抗体、钠离子不依赖中性氨基酸转运蛋白LAT1抗体、Y+系统阳离子氨基酸转运蛋白抗体或CD98LC抗体),可通过WB、IP、IF和ELISA方法检测来自小鼠、大鼠和人类的LAT1蛋白。LAT1抗体(D-10)以非偶联型抗LAT1抗体形式提供,同时也有多种偶联形式的抗LAT1抗体,包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。L型氨基酸转运蛋白1(LAT1)是一种多通道膜蛋白,负责钠离子不依赖的大型中性氨基酸的高亲和力转运。LAT1与氨基酸转运蛋白CD98以二硫键连接形成异二聚体而发挥作用。LAT1主要在成人肺和肝脏中表达,但也表达于脑、胸腺、视网膜、睾丸、胎盘、骨髓和胎儿肝脏中。在视网膜中,LAT1定位于血视网膜屏障(BRB),介导L-亮氨酸从血液到视网膜的转运。苯丙酮尿症对大脑的毁灭性影响是由于血液中高浓度的苯丙氨酸导致血脑屏障上LAT1水平升高所致。LAT1接受与氨基酸相关的抗癌药物美法仑,并在食管鳞状细胞癌的细胞增殖、分化和侵袭中发挥重要作用。

Good afternoon, I was wondering if you have any insights on detecting the LAT1 protein using your antibody by Western blot in RIPA-extracted mouse cardiac tissue, specifically around 25KDa?

Thank you for your question. LAT1 Antibody (D-10): sc-374232 was tested and is recommended for detection of LAT1 of any mouse, rat, or human cell or tissue sample expressing the protein, by WB, IP, IF and ELISA. Observed molecular weight of LAT1 is 40 kDa; molecular weight of glycosylated LAT1 is 45-55 kDa.

Hello, which secondary antibodies would you recommend for IHC with this primary antibody? Which blocking agent would you recommend to use for IHC? Thank you

Thank you for your question. For HRP-based secondary detection of Anti-LAT1 Antibody (D-10): sc-374232 we recommend m-IgG1 BP-HRP: sc-525408. Our plant-based UltraCruz® Blocking Reagent: sc-516214 is recommended for blocking in IHC applications.

Hi, if this LAT1 antibody in conjugation with HRP is recommend by you for IHC(P) applications (citation: "HRP für WB, IHC(P) und ELISA"), which antigen retrieval method to your knowledge have been tested successfully for this antibody?

Thank you for your question. The HRP conjugated primary antibody is recommended and warrantied for immunohistochemistry on formalin-fixed paraffin-embedded tissues, or for IHC(P), however we are still gathering data to show on our website. Check back for updates! In general, we recommend using a sodium citrate buffer (pH 6.0) with heat treatment for antigen retrieval.

I am interested in using this anti-body in a porcine model. Is there any data showing that the reagent can detect porcine?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. Rated 1 out of 5 by from Variable/weak wb results from mouse CNS tissuesI've tried to use this antibody to detect Lat1 of mouse origin in cell lines and tissues from the brain and spinal cord, and I get variable signal. The protein is known to be expressed in my cells of interest, and the mRNA is really quite high as well. Either this antibody is less efficient for Ms Lat1, or its not specific. Based on this I wouldn't trust it for ICC unless using an over-expression model.
