Laminin α-4 Antibody (6C3) 是一种IgG1 κ小鼠单克隆Laminin alpha-4抗体（也称为LAMA4抗体或Laminin亚基alpha-4抗体），可通过WB、IP和ELISA检测人源Laminin alpha-4蛋白。Laminin α-4 Antibody (6C3)可作为非偶联的抗Laminin alpha-4抗体使用。层粘连蛋白是定位于基底膜的重要且丰富的结构性非胶原糖蛋白。基底膜（细胞相关的细胞外基质(ECMs)）是层粘连蛋白的聚合物，具有稳定的IV型胶原网络、巢蛋白和几种蛋白聚糖。基底膜位于上皮层下方，血管内皮周围，以及肌肉、外周神经和脂肪细胞周围。基底膜的形成影响细胞增殖、表型、迁移、基因表达和组织结构。每种层粘连蛋白都是由 α, β, and γ 链亚基组成的异三聚体，这些亚基经历细胞分泌并整合到ECM中。层粘连蛋白可以自我组装，与其他基质大分子结合，并通过整合素、抗肌萎缩蛋白聚糖和同源层粘连蛋白受体介导独特和共享的细胞相互作用。人类层粘连蛋白α-4基因定位于6号染色体q21区，并在成人心脏、肺、卵巢、小肠和大肠、肝脏和胎盘中表达。

For Western Blot, is it recommended to use denatured or non-denatured conditions with Laminin α-4 (6C3): sc-130541 antibody?

Rated 4 out of 5 by from Staining of human RCC paraffin embedded tissueThe antibody works well on human paraffin sections of renal cell carcinoma. Matching normal kidney tissue of the same RCC patient shows some Lama4 staining. Protocol After deparaffinization with xylene and ethanol block endogenous peroxidase with 0.3% hydrogen peroxidase/PBS 15 min at RT. Antigen retrieval with 10mM citrate buffer pH 6.0, block 5% BSA/PBS 1h at RT, anti-lama4 Ab dilution 1:30 in 0.5% BSA/PBS O.N. at 4C, 2nd Ab goat anti-mouse biotin (Dako, ref E0433) diution 1:500 in 0.5% BSA/PBS 45 min at RT. Strep-HRP (Dako, ref P0397) 1:200 in 0.5% BSA/PBS 30 min at RT. Develop with DAB, counterstain with Mayer's hematoxylin.
