INSM1抗体（A-8）是一种IgG1 κ小鼠单克隆INSM1抗体（也称为INSM1抗体），可通过WB、IP、IF、IHC(P)和ELISA检测到小鼠、大鼠和人源的INSM1蛋白。INSM1抗体（A-8）既有非偶联形式的抗INSM1抗体，也有多种偶联形式的抗INSM1抗体，包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。INSM1（胰岛素瘤相关蛋白1），也称为锌指蛋白IA-1，是一种发育调控的锌指转录因子。它定位于细胞核内，并在经历神经内分泌分化的胚胎组织中表达。INSM1在正常成人组织中不表达，但在神经内分泌肿瘤中高表达。INSM1包含5个Cys(2)-His(2)型锌指DNA结合域和一个前激素域。INSM1作为Neuro D启动子的转录抑制因子，并招募细胞周期蛋白D1作为共抑制因子。它在神经内分泌发育中起重要作用，是胰腺内分泌细胞正常分化所必需的。抑制INSM1会导致胰高血糖素和胰岛素阳性细胞的形成减少。编码INSM1的基因直接受Neurogenin 3调控，Neurogenin 3与INSM1启动子区域的染色质结合并诱导转录。

Thank you for your question. We have not validated the antibody for canine. The antibody is recommended for mouse, rat and human samples.

Thank you for your question. If your samples are PFA-fixed and/or paraffin-embedded, antigen retrieval is advised. You can find details in our Immunoperoxidase Staining protocol: https://www.scbt.com/resources/protocols/immunoperoxidase-staining Here is the recommended method, which is heat treatment: Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

Rated 3 out of 5 by from サンプル量がやや少なかったですが、概ね良好そうでした。サンプルを50µlいただいたのですが、50倍希釈を予定していた（他院の条件を参照した）が、BOND-Maxでは量が足りなかったため80倍希釈のER1（pH6.0）で20分賦活しました。抗体反応時間は15分です。結果は過染気味でしたが標的の核は良好な染色強度でした。希釈範囲が50-500であったので、さらに希釈すれば良好な結果になりそうです。
