Id4抗体（B-5）是一种IgG1 κ小鼠单克隆Id4抗体（也称为ID4抗体或DNA结合蛋白抑制剂ID-4抗体），可通过WB、IP、IF和ELISA检测小鼠、大鼠和人源的Id4蛋白。Id4抗体（B-5）既可以是非偶联的抗Id4抗体形式，也可以是多种偶联形式的抗Id4抗体，包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。基本的螺旋-环-螺旋（bHLH）蛋白的Id家族成员包括Id1、Id2、Id3和Id4。它们普遍表达并与A类和B类HLH蛋白的成员二聚化。由于缺少基本区域，因此所得的异二聚体不能结合DNA。因此，Id型蛋白似乎负调节bHLH蛋白的DNA结合。由于Id1抑制E12和Myo D的DNA结合，因此它显然起抑制肌肉特异性基因表达的作用。在促进肌肉细胞分化的条件下，Id蛋白水平下降，允许E12和/或E47与Myo D和肌细胞生成素形成异二聚体，从而激活肌源性分化。已经显示，每种Id蛋白的表达都强烈依赖于生长因子的激活，并且通过反义寡核苷酸降低Id mRNA水平会导致生长因子刺激后停滞的细胞延迟重新进入细胞周期。

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Id4 (B-5): sc-365656 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 5 out of 5 by from Id4 antibody (B-5) works great for IF stainingI tried the Id4 antibody (B-5) on both 293T cells and human Id4 transfected 293T cells for the immunofluorescence staining at 1:300. It gave very specific nuclear staining on the endogenous Id4 in non-transfected 293T cells. For the human Id4 transfected 293T cells, the Id4 shows both cytoplasmic and nuclear staining pattern, which is consistent with the published data reporting Id4 cytoplasmic and nuclear localization in prostate cancer cells.

Taking the time to try this antibody, found good effect in 293T cells: closed fast closed closed 30min beyotime liquid, first antibody is the product (1:700 dilution), second antibogy buy from abclonal anti mouse (1:30000 dilution), the exposure time is 60 seconds.The molecular weight is about 18KD.But there was no band on the rat BMSC cells, and that was the case for the two time. It is also necessary to continue to work hard.The picture is the result of 293T cell samples

Rated 1 out of 5 by from only background, no bands at expected size in WBIn a large panel of cell lines expressing high levels of ID4 we didn't get a specific signal at the expected protein size of 18kDa, however the overall background was quite strong. We used the antibody in TBST, 5% BSA diluted 1:100.
