Geminin抗体（F-7）是一种小鼠单克隆IgG2b kappa轻链抗体，可通过蛋白质印迹（WB）、免疫沉淀（IP）、免疫荧光（IF）、免疫组织化学和酶联免疫吸附测定（ELISA）检测小鼠、大鼠和人源性Geminin蛋白 ）。抗Geminin抗体（F-7）有非结合型和多种结合型，包括琼脂糖、辣根过氧化物酶（HRP）、藻红蛋白（PE）、异硫氰酸荧光素（FITC）和多种Alexa Fluor®结合物。Geminin在细胞周期中调节DNA复制起始方面起着至关重要的作用，确保DNA在每个周期中仅复制一次，以保持基因组的稳定性。Geminin (F-7) 抗体可抑制小染色体维持（MCM）蛋白组装成复制前复合物，从而防止过早的DNA复制。在整个细胞周期中，Geminin的水平受到严格调控，在G1期达到最低，在S、G2和M期达到最高。DNA复制起始所必需的Geminin降解在细胞分裂过程中通过APC介导的泛素蛋白水解发生。这种对Geminin的精确调节可维持细胞功能，因为失调会导致基因组不稳定并促进肿瘤发生。

What would be a good secondary antibody AF647 for SC-74456? Is there IgG2 specific secondary

Thank you for your question. We would recommend using our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to CruzFluor™ 647 (CFL 647), m-IgGκ BP-CFL 647: sc-516179. This is our strongly recommended alternative to conventional anti-mouse IgG secondary antibodies for RGB Western blotting (WB [RGB]), Immunofluorescence (IF) and Flow Cytometry (FCM). We do not currently offer an anti-mouse IgG2 isotype-specific secondary antibody at this time.

I ordered Geminin (F-7): sc-74456 for IHC purpose in research work on human paraffin sections, its data sheet which tells that the concentration is 1:50-1:500, the vial size is 200 ug/ml ..I need to know how many slides does this amount suffice

Thank you for your question. You can find our recommended protocol for indirect immunoperoxidase staining under this link: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining. A primary antibody solution of approximately 100 µl per slide is usually adequate. At a dilution of 1:50, one ml of antibody solution is sufficient for 500 slides, or the tenfold amount (5000 slides) if a dilution of 1:500 is sufficient.

I am using Geminin (F-7): sc-74456 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
