I have two questions about this antibody.1)The two cell lines that you show (HeLa and Mia PACA) were transfected with glucokinase or this is an endogenous expression? 2) is HepG2 a positive control?

Thank you for your question. The immunofluorescence data for this antiobody is in endogenous samples. HepG2 is one of the recommended positive controls for this protein.

I wonder whether this antibody has ever been used to detect GCK in tumor tissues other than liver. Do you have any evidence that this protein is expressed also in cancers other than hepatocarcinomas?

GCK (glucokinase, NP_000153.1) does not expresses in HepG2. Besides, GCK MW=52kDa and in the band shown in the wb migrates quite higher. Has the identity of the shown band been verified by sequencing?.

Thank you for your question. There is literature supporting expression of GCK in liver. If you would like to share with us any literature reference supporting a lack of expression of GCK in the HepG2 cell line we are happy to review the reference and reevaluate our data. We have not verified the identity of the bands detected in Western with this antibody through sequencing. Using this mouse monoclonal antibody, GCK (G-6): sc-17819, we have observed GCK at a molecular weight higher than the predicted molecular weight of 52 kDa in multiple cell lines. This difference may be due to post-translational modifications of the protein, or other factors which would influence the observed molecular weight of the protein in an SDS-PAGE gel.

What is the recommended fixation method for immunofluorescence staining with GCK (G-6): sc-17819 monoclonal antibody?

Rated 4 out of 5 by from Two bandsPerformed immunoblotting with this antibody at 1:500 in MEF whole cell lysates with RIPA. I saw a band at around 50 kD and another at around 80-90 kD. The sample pictures shown for this antibody looks like the band is between 55 and 90 kD, while the predicted band size from the specification sheet says 50 kD. So its confusing as to which band should be used.

Rated 5 out of 5 by from Good resultThis antibody is used for evaluating activity of the GCK after stimulation or inhibition of GCK in lung cancer cell lines. The antibody is very reliable. We have found that dilution in 5 % BSA is optimal .Dilution: 1:500 is good choice dilution.
