CUL-2抗体(C-4)是一种IgG1 κ型小鼠CUL-2单克隆抗体(又称Cullin 2抗体或CUL2抗体),通过WB、IP、IF和ELISA检测小鼠、大鼠和人源的CUL-2蛋白。CUL-2抗体(C-4)既可以是非偶联的抗CUL-2抗体形式,也可以是多种偶联的抗CUL-2抗体形式,包括琼脂糖,HRP, PE, FITC和多个Alexa Fluor®偶联物。Cullin蛋白包含一个独特的介质家族,参与蛋白的选择性靶向泛素(Ub)介导的蛋白水解。CUL-1是酵母Cdc53的哺乳动物同源物,是E3泛素连接酶复合物SCF的组成部分。SCF (Skp1/CUL-1/F-box蛋白复合物)由Skp1和CUL-1以及F-box蛋白(如Skp2)组成,后者决定了复合物的底物特异性。cul -1介导的泛素化导致细胞周期蛋白cyclin D, p21和cyclin E的降解。另一种cullin, CUL-3,促进了不依赖于SCF活性的cyclin E的降解,而CUL-2与肿瘤抑制蛋白VHL和延长蛋白B结合形成VBC复合物,其结构类似于SCF连接酶。蛋白质水解也通过CUL-4与Nedd-8(一种泛素样蛋白)结合的方式发生,在那里它也作为多功能E3复合体的活性成分发挥作用。CUL-5也被称为血管加压素激活的钙动员蛋白(vcam -1),因为它与CUL-1具有显著的序列同源性,因此也包括在cullin家族中。
