What is the sequence of the target epitope for this antibody? Is it in the extracellular region of CD28?

Thank you for your question. This mouse monoclonal antibody, CD28 (CD28.2): sc-19655, is raised against human CD28 transfected murine T cell hybridoma. The exact epitope recognized by the antibody has not been determined. We do not know whether the epitope is extracellular or intracellular for this clone.

Can it be uesd to active T cells for proliferation?

Thank you for your question! Our labs have not tested this antibody clone regarding the possibility to activate T-cell proliferation. However, this clone has been published for the use in functional studies on human Samples. You might be able to find further information here: Chen ML et al. Antipsychotic drugs suppress the AKT/NF-kB pathway and regulate the differentiation of T-cell subsets. Immunol Lett 140:81-91 (2011) If you were to test this antibody in functional studies, we suggest to use this antibody provided in an azide-free (Low endotoxin level) buffer. Can CD28 (CD28.2): sc-19655 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD28 (CD28.2): sc-19655 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
