I am wondering if we can detect both pro and cleaved caspase 1 with this sc-392736 antibody as I always get two bands every time with 1: 750 dilution.

What is the difference between D-3 and D-4? Do these detect cleaved caspase 1 as well?

Thank you for your question. These are both mouse monoclonal antibodies, but they are have different isotypes, and are raised against different antigens. The caspase-1 Antibody (D-3): sc-392736 is raised against an epitope mapping between amino acids 367-391 (human origin), and caspase-1 p20 Antibody (D-4): sc-398715 is raised against an epitope mapping between amino acids 259-294 (mouse origin). Both antibodies are expected to detect cleaved caspase, however (D-3) should recognize the p10 subunit, and (D-4) should recognize the p20 subunit. Please contact our Technical Service Department for help in choosing the best antibody for your research.

I used SC-1780 (caspase-1 Ab) to detect cleaved-caspase-1 (p20) polyclonal antibody. I would like to change the Ab to SC-392736 monoclonal Ab. But there is not any comment for detection of cleaved-caspase-1 (p20) protein. Can I detect using SC-392736?

Thank you for your question. We would not recommend this mouse monoclonal, caspase-1 Antibody (D-3): sc-392736, for detection of the cleaved caspase-1 p20 subunit. This antibody is specific for an epitope mapping between amino acids 367-391 near the C-terminus of caspase-1 of human origin. And according to Uniprot, the p20 subunit corresponds to a different region between amino acids 120-297. http://www.uniprot.org/uniprot/P29466#ptm_processing
