αPAK抗体(A-6)是一种IgG1 κ小鼠单克隆αPAK抗体,可通过WB、IP、IF和ELISA检测小鼠、大鼠和人类来源的αPAK、βPAK和γPAK。αPAK抗体(A-6)可作为非偶联的抗αPAK抗体形式,也可作为多种偶联形式的抗αPAK抗体,包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。已显示三种丝氨酸/苏氨酸激酶异构体,即αPAK p68、βPAK p65和γPAK p62,与参与信息素信号传导的酿酒酵母激酶Ste 20具有高度序列同源性。α、β和γPAK异构体在其活性GTP结合状态下特异性地与Rac1和Cdc42结合,抑制其内在的GTP酶活性,导致其自磷酸化。γPAK上有八个自磷酸化位点,包括Ser 19、Ser 141和Thr 402,而Ser 141和Thr 402的磷酸化与γPAK的激活相关。一旦磷酸化且其对Rac/Cdc42的亲和力降低,PAK异构体就会从复合物中解离出来寻找下游底物。其中一个这样的假定底物是Mek激酶,它是Mek4的上游效应器,参与JNK信号通路。虽然PAK异构体以GTP依赖的方式与Rac1和Cdc42相互作用,但它们不与Rho相互作用。
