O anticorpo PDGFR-α (C-9) é um anticorpo PDGFR-alfa monoclonal de camundongo IgG1 κ (também designado anticorpo PDGFRA ou anticorpo CD140a) que detecta a proteína PDGFR-alfa de origem de camundongo, rato e humana por WB, IP, IF, IHC(P) e ELISA. O anticorpo PDGFR-α (C-9) está disponível na forma de anticorpo anti-PDGFR-alfa não conjugado, bem como em várias formas conjugadas de anticorpo anti-PDGFR-alfa, incluindo agarose, HRP, PE, FITC e vários conjugados Alexa Fluor®. O fator de crescimento derivado de plaquetas (PDGF) é um mitógeno para células derivadas do mesênquima e da glia. O PDGF consiste em duas cadeias, A e B, que se dimerizam para formar isoformas funcionalmente distintas: PGDF-AA, PDGF-AB e PDGF-BB. Essas três isoformas se ligam com afinidades diferentes a dois tipos de receptores, PDGFR-α e -β, que são dotados de domínios de proteína tirosina quinase. O PDGFR-α pode se ligar às subunidades A e B do PDGF, enquanto o PDGFR-β pode se ligar apenas à subunidade B. A ligação do fixador promove a homo ou heterodimerização dos receptores de PDGF de uma maneira específica. O PDGF-AA induz a dimerização de dois receptores α, o PDGF-AB induz a dimerização de αα e αβ e o PDGF-BB induz a formação de três tipos de dímeros, αα, αβ e ββ. Os genes que codificam o PDGFR-α e -β estão localizados no cromossomo humano 4q12 e 5q33.1, respectivamente. A translocação do gene PDGFR-β com o gene TEL está associada à leucemia mielomonocítica crônica (CMML), uma síndrome mielodisplásica, e demonstra o potencial oncogênico dos receptores PDGF.

Thank you for your question. We do not have internal image data for flow cytometry use of this antibody yet, but the antibody was used in FCM in publication PMID 30057110.

Hi, I was wondering if this antibody has been tested to cross-react with PDGFRbeta? We would like to buy a highly specific PDGFRalpha antibody for use on a Western blot.

Thank you for your question. The antibody was raised against amino acids 951-1089 of PDGFR- of human origin with accession number https://www.ncbi.nlm.nih.gov/protein/129892 The exact binding region of this antibody within the antigen has not been mapped. This antibody has not been tested experimentally for potential cross-reactivity under denaturing conditions in a Western Blot against different proteins. You might want to align the antigen sequence against the protein form of interest in order to determine potential reactivity. Hi, can you please advise if for staining on cells it is necessary to permeabilise cells with Triton-X? Is the antibody binding to an intra- or extra-cellular site? Many thanks in advance.

Thank you for your question. PDGFR-α Antibody (C-9) is a mouse monoclonal IgG1 raised against amino acids 951-1089 of PDGFR-α of human origin. According to UniProt, this corresponds with a cytoplasmic region of the protein (https://www.ncbi.nlm.nih.gov/protein/P16234). If you are using a formalin-based fixative (e.g., PFA) then we do recommend including a cell permeabilization treatment. After fixing the cells, wash in three changes of PBS. Then incubate for 3 minutes in a solution of .01% Triton X-100 in PBS. Followed by a wash in three changes of PBS. L'anticorps monoclonal PDGFR-α (C-9) sc-398206 a été élaboré contre les acides aminés 951-1089 de PDGFR-α d'origine humaine, correspondant au numéro d'accession NCBI P16234. https://www.ncbi.nlm.nih.gov/protein/P16234 L'anticorps est recommandé pour la détection de la protéine humaine, de souris, de rat, équine, canine, bovine et porcine. Notre laboratoire n'a malheureusement pas testé la réactivité avec PDGFR-α de poulet (aviaire). L'homologie entre les séquences des protéines de poulet et humaine est relativement élevée et il est très probable que l'anticorps réagisse aussi avec PDGFR-α aviaire.
