HPS-1 Antibody (5G12G2) is an IgG2a κ mouse monoclonal HPS-1 antibody (also designated HPS-1 antibody) suitable for the detection of the HPS-1 protein of mouse, rat and human origin by WB, IP and ELISA. HPS-1 Antibody (5G12G2) is available as both the non-conjugated anti-HPS-1 antibody form, as well as multiple conjugated forms of anti-HPS-1 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Hermansky-Pudlak syndrome (HPS) is a rare, genetically heterogeneous, autosomal recessive disorder. It is characterized by oculocutaneous albinism, lysosomal storage defects and prolonged bleeding due to platelet storage pool deficiency. There are 10 HPS genes encoding HPS proteins that all interact within three distinct ubiquitously expressed protein complexes or biogenesis of lysosome-related organelle complexes. Defects in these genes cause HPS. HSP-1 is a component of multiple cytoplasmic organelles and is important for their normal development and function. It plays a role in intracellular protein sorting.

I am planning to perform a CRISPR K.O. cell line by NHEJ and Cas9n. I would like to do a WB to confirm my cell line does not produce functional protein. Thus I wonder where this antibody binds.

Thank you for your question. The epitope has not been mapped for this antibody, so we are unable to provide the exact antibody binding site. However, this will be fine to use for your purpose with CRISPR.

For Western Blot, is it recommended to use denatured or non-denatured conditions with HPS-1 (5G12G2): sc-101435 monoclonal antibody?
