FBXO42 Antibody (FL-6) is an IgG_2a κ mouse monoclonal FBXO42 antibody (also designated FBXO42 antibody) that detects the FBXO42 protein of mouse, rat and human origin by WB, IP and ELISA. FBXO42 Antibody (FL-6) is available as the non-conjugated anti-FBXO42 antibody. FBXO42 (F-box only protein 42), also known as FBX42, is a 717 amino acid protein that contains four Kelch repeats and one N-terminal F-box domain, and belongs to the F-box family of proteins. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein) type E3 ubiquitin ligase complex and are involved in substrate recognition and recruitment for ubiquitination. They are members of a larger family of proteins that are involved in the regulation of a wide variety of cellular processes (including the cell cycle, immune response, signaling cascades and developmental processes) through the targeting of proteins, such as cyclins, cyclin-dependent kinase inhibitors, IkB-a and b-catenin, for degradation by the proteasome after ubiquitination. Expressed at moderate levels in liver, ovary, kidney, testis and adult brain, and at low levels in heart and fetal brain, FBXO42 directly interacts with Skp1 p19 and CUL-1.

For Western Blot, is it recommended to use denatured or non-denatured conditions with FBXO42 (FL-6): sc-100737 antibody?

Thank you for your question. We recommend this antibody for use in denatured Western Blot conditions. It has not been validated for use in non-denatured conditions. Please contact our Technical Service Department for further details or inquiries.

Rated 4 out of 5 by lyq612 from Works well for WB in PANC1 line. Distinct band observed, though also mixed with a ghost non-specific band at ~75 KDa. Tested FBXO42 shRNA knockdown, CRISPR-knockout (homozygous frameshift deletion), and FBXO42-overexpressed (via Lentivirus) clonal stable PANC1 lines. Used this Ab (Lot #: B1220) for WB (1:800 in 5% milk-TBST0.1%, 0.125 ug/mL final, overnight ~10 hours) to test PANC1 cell lysates. 20 ug total proteins of cell lysates were each resolved on a Tris-Glycin gel (4-12% gradient) and transferred to a PVDF membrane. Antibody was freshly diluted in milk without freeze-n-thawing.
