CYP8B1 Antibody (M15-P3B7) is a high quality monoclonal CYP8B1 antibody (also designated CYP8B1 antibody) suitable for the detection of the CYP8B1 protein of mouse, rat and human origin. CYP8B1 Antibody (M15-P3B7) is available as both the non-conjugated anti-CYP8B1 antibody form, as well as multiple conjugated forms of anti-CYP8B1 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. CYP8B1 (sterol 12-alpha-hydroxylase) is a member of the cytochrome P450 superfamily of monooxygenase enzymes that are involved in the metabolism of a wide array of endogenous and xenobiotic compounds. CYP8B1 is highly expressed in liver and is an important enzyme for bile acid synthesis. Specifically, CYP8B1 moderates the ratio of cholic acid over chenodeoxycholic acid to control the solubility of cholesterol. The gene encoding human CYP8B1 maps to chromosome 3p22.1. The CYP8B1 gene encodes a 501-amino acid protein and does not contain any introns. The CYP8B1 gene promoter is transactivated by hepatocyte nuclear factor 4a. In mice, disruption of the CYP8B1 gene prevents the synthesis of cholate, which is a primary bile acid.

Are there any issues with using sc-101387: CYP8B1 (M15-P3B7) mouse monoclonal primary antibody on mouse samples?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody. Unspecific bandsWe got CYP8b1 and FGFR4 antibody samples and they do not work for WB application. For both these antibodies tried on murine liver samples (on different blots) we got unspecific bands at exact same MW which we could not get rid off even after harsh stripping. It took us lot of time and energy for nothing in the end.

Cyp8b1 antibody gives good bands on Liver WesternI received Cyp8b1 sample antibody with Cyp27a1 sample antibody and both gave good bands on mouse liver westerns. Used 1:1000 dilutions for primary and secondary antibodies, and "femto" enhancer instead of regular "pico" enhancer. Got really good bands, see picture

WB ResultsI applied for a sample two week ago.My sample is the mouse liver. Incubated the antibody for overnight at 4℃, 1:1000 dilution, and m-IgGκ BP-HRP incubated for 1 hour at RT , 1:1000 dilution.The results are shown in the figure.
